Depends on how much you want to spend. Yarn.com can have some great bargains and a huge selection. Knitpicks is okay, but working with their Felici, I keep feeling as if it's going to have no elasticity in my socks (not that I'm going to be wearing my self-striping Fourth Doctor socks to the office on a regular basis.
I shop the The Loopy Ewe, but I've also been shopping the dyers themselves. The Verdant Gryphon only has purples up at the moment, but they're just restarting after the two halves of The Sanguine Gryphon splitting. Their Eidos yarn is very yummy, though. There's Indigodragonfly, which has a decidedly fannish, silly bent with names such as "The Earth Is Doomed. (Giles)", "Only, The Exact Phrase I Used Was, “Don’t.” (Inara)", and "Bright Lights, Big City, Busy Highway, Slow Unicorn." Plus, they custom dye any of their standard colors, so if you want, say, three skeins of Self Elmolation for a project, you can pick your base and order them dyed up.
My current favorite yarn shopping, though, happens on Ravelry, seeing what people have in their stashes for sale or trade. You can spend hours browsing that.
